I just finished Telltale's The Walking Dead Season 2, and Kenny is one of my favorite characters of all time in any video game. He's a rough one to get along with at timea, but if you side with him, you'll get rewarded much more than with any other character in that game (any most characters from any game). The ending of S2 with Kenny (optional) is one of my favorite endings ever in a video game, despite the gameplay being boring.
As for other characters, there are of course Wrex and Garrus from Mass Effect. I almost exclusively just pick those two in the first Mass Effect. The comradery between them and Shepard is unopposed from any other characters in the series. Just the soothing way Wrex calls out Shepard is awesome. And what I love about Mass Effect as a series, is that if you put effort into characters, you'll get rewarded for it.
So what are some of yours favorite supporting characters in video games?

I like the entire supporting team in Metal Gear Solid. Of course, the great voice acting is part of it. I also quite like the dog and Quiet in MGS V. They're helpful, one of them is a nice doggie, the other is... well, it's Quiet, wootwoot. Rush in the Mega Man series. It's quite a simple idea: instead of just getting the transport items, like in Mega Man 2, have a companion dog transform into them. It gave so much personality to the game. Alyx in Half-Life 2 and the Episodes is one of the most helpful, well-written and well-acted characters I can think off, which is a good thing, as she is by your side through the entirety of Ep. 1. The bird from Astal (hey, look, Spirit Bomb! ). Besides being very helpful, the reveal at the end gives its actions even more meaning. The entire supporting cast from Chrono Trigger. Man, that game is just something else. Talk about a perfectly likeable bunch of characters.

Luigi hands down, best supporting character ever, a taller clone in green that jumps higher, it works perfectly.

Almost anyone from Mass Effect 2 depending on how you play the game. Jack and Jacob are pretty boring though. Jack works great if she is a love interest. Otherwise you don't get her full story arc. Jacob is the reserved stoic. Take it for what he is.

This is a hard one. Cortona is pretty damn good, if you ask me. I'd also say Yoshi is a great choice as well. I guess the best long term supporting character of all time has to be the Chocobo. The constant support the Chocobo has brought to the Final Fantasy series can only be compared to Luigi in the Mario series.

The horse from Shadow of the Colossus, Agro. You really feel for that horse when it falls down during the last Colossi, and the main characters shouts his name. Sad, so sad.
